Tomographic acquisition in TEM

It is possible to perform 3D characterisation of thin cut samples out of 2D conventional TEM imaging. Once an area of interest is located, a) electron projection images are collected with an as high as possible tilt range with a certain angular step (for example between  ± 65° and 1° step), and then b) the obtained tilt image series is used to reconstruct a three-dimensional image of the targeted area.  

Tomographic reconstruction

To reconstruct the imaged area from the tilt series of projection images, dedicated software and algorithms are needed as well as a visualisation toolkit. 

Here on the side shows an example of a reconstructed volume rendered in 3D from a biological sample.
